By locking the seam of BamA, I aim to indentify the mechanism by which the BAM complex is able to insert nascent outer membrane proteins (OMPs). Using both听in vivo听补苍诲听in vitro听assays, I can measure the activity of BamA and how locking the barrel effects that activity. The aim of this project is to identify if BamA uses lateral gate opening to directly pass nascent OMPs into the membrane.听
